Target Background

Function
Lipid transporter involved in lipid countertransport between the Golgi complex and membranes of the endoplasmic reticulum: specifically exchanges sterol with phosphatidylinositol 4-phosphate (PI4P), delivering sterol to the Golgi in exchange for PI4P, which is degraded by the SAC1 phosphatase in the endoplasmic reticulum. Displays a similar affinity for PI4P and sterols. Binds sterol and PI4P in a mutually exclusive manner (Probable).
Gene References into Functions
  1. the sterol content in the mitochondrial fraction was significantly decreased in vivo after Osh4 inactivation in a genetic background in which all the other OSH genes were deleted. PMID: 29487131
  2. Osh4-mediated exocytosis depends upon lipid binding and exchange without a strict requirement for sterol. PMID: 28993464
  3. Osh4p transports sterol against its gradient between two membranes by dissipating the energy of a phosphatidylinositol-4-phosphate gradient. PMID: 25849868
  4. Osh4p is needed to reduce the level of phosphatidylinositol-4-phosphate on secretory vesicles as they mature. PMID: 25165144
  5. X-ray crystallographic determination of the conformational changes Osh4 undergoes during ligand binding. PMID: 23756172
  6. Kes1 (Osh4p) associated with exocytic vesicles that move from the mother cell into the bud. PMID: 21819498
  7. Study reports that Kes1 integrates multiple aspects of lipid metabolism in distal stages of the secretory pathway with TORC1 and nitrogen signaling. PMID: 22341443
  8. The structure of the Osh4p-phosphatidylinositol 4-phosphate complex was solved. It was shown that Osh4p quickly exchanges dehydroergosterol for phosphatidylinositol 4-phosphate. PMID: 22162133
  9. structure of the open state of a lid-truncated form of Osh4 was determined at 2.5 A resolution PMID: 16136145
  10. Osh4p/Kes1p, specifically facilitates the nonvesicular transfer of cholesterol and ergosterol between the plasma membrane and endoplasmic reticulum in vitro PMID: 16585271
  11. Kes1p regulates Golgi apparatus phosphatidylinositol-4-phosphate function PMID: 17881569
  12. The mechanism used by Osh4 to release cholesterol is conceptualized as a molecular ladder with the rungs being amino acids or water-mediated amino acids that interact with 3-hydroxyl group of cholesterol PMID: 18937371
