Target Background

Function
Recruits UPF2 at the cytoplasmic side of the nuclear envelope and the subsequent formation of an UPF1-UPF2-UPF3 surveillance complex (including UPF1 bound to release factors at the stalled ribosome) is believed to activate NMD. Binds spliced mRNA upstream of exon-exon junctions. Involved in nonsense-mediated decay (NMD) of mRNAs containing premature stop codons (premature termination codon PTC) by associating with the nuclear exon junction complex (EJC) and serving as link between the EJC core and NMD machinery. Eliminates the production of nonsense-containing RNAs (ncRNAs). Required for plant development and adaptation to environmental stresses, including plant defense and response to wounding.
Gene References into Functions
  1. The results demonstrate that, in contrast to mammalian cells, a delicate balance of UPF3 transcript levels by its feedback loop and by restriction of its transcription, are crucial for proper nonsense-mediated-decay regulation in Arabidopsis. PMID: 25820429
  2. Here, we analyzed the abundances of all splice variants for each alternatively spliced gene in an Arabidopsis mutant that lacks UPF3, one of the core components of NMD machinery, to determine if the PTC-containing transcripts are degraded by NMD. PMID: 19863731
  3. Our results indicate that the AtUPF3 gene is required by the plant nonsense-mediated mRNA decay system to obviate aberrantly spliced mRNA. PMID: 16098107
  4. Data show that in mutants impaired in the nonsense-mediated decay (NMD) components UPF3, levels of the alternatively spliced AtGRP7 and AtGRP8 transcripts are strongly elevated, implicating UPF3 in the decay of these clock-regulated transcripts. PMID: 17924945
  5. The nonsense-mediated decay factor UPF3 localize to the nucleolus, suggesting that the Arabidopsis nucleolus is therefore involved in identifying aberrant mRNAs and NMD. PMID: 19602621
