Target Background

Function
Protein kinase that plays a central role in centrosome duplication, control of centrosome size, spindle formation and nuclear envelope breakdown during cell divisions. Paternal copy is required to regulate synthesis of daughter centrioles prior to fertilization. Maternal copy regulates centrosome duplication during later cell cycles. Functions upstream of sas-5 and sas-6, and is required for their localization to the centrosome. Its role in nuclear envelope breakdown is mediated by the spindly-like protein spdl-1 and the RZZ complex, which in turn recruits the spindle checkpoint proteins mdf-1 and mdf-2, dynein and dynactin to unattached kinetochores.
Gene References into Functions
  1. These findings lead us to conclude that ZYG-1 promotes limited centriole amplification solely during the symmetric division in the C. elegans seam lineage. PMID: 29307730
  2. Structure of the C. elegans ZYG-1 cryptic polo box suggests a conserved mechanism for centriolar docking of Plk4 kinases. PMID: 24980795
  3. Direct interactions of the SAS-6 coiled coil with ZYG-1 and SAS-5 are required to recruit SAS-6 to the mother centriole. PMID: 23673331
  4. Data show that levels of the C. elegans Plk4 homolog ZYG-1 are elevated by impairing proteasome or SCF function, indicating that ZYG-1 is regulated by a conserved mechanism. PMID: 22623721
  5. Data reveal differences in the regulation of mitotic and meiotic centrosome duplication, particularly with regard to ZYG-1 activity, and reveal an important role for centrosomes in spermatid formation. PMID: 20144993
  6. Study demonstrates that the kinase ZYG-1 phosphorylates the coiled-coil protein SAS-6 at serine 123 in vitro; this phosphorylation event is crucial for centriole formation in vivo. PMID: 20059959
  7. Numerous candidate regulators of centrosome duplication and regulatory mechanism involving factors that tether the centrosome to the nucleus, have been identified. PMID: 17446307

Subcellular Location
Cytoplasm, cytoskeleton, microtubule organizing center, centrosome. Cytoplasm, cytoskeleton, microtubule organizing center, centrosome, centriole. Note=Component of the centrosome (PubMed:11371350). Present at centrioles only immediately before their duplication (PubMed:11371350). Expression at centrosomes increases from prophase to anaphase during mitosis (PubMed:22623721).
