Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Notes

Target Background

Function
Required for normal accumulation and movement of lipid-modified hedgehog (hh) morphogen. May act by stabilizing the interaction between heparan sulfate proteoglycans (HSPGs) and hh, HSPGs being required for diffusion of hh morphogen. Not involved in wingless (wg) morphogen movement, suggesting that it may provide HSPG specificity for Hh.

  2. study concludes that the WIF domain is responsible for the divergence between Drosophila Shf and human Wif-1, channeling the recognition either toward Hh or Wg signal, whereas the EGF repeats give the protein the ability to interact with glypicans PMID: 22951645
  3. Shf is the ortholog of the human Wnt inhibitory factor (WIF), a secreted antagonist of the Wingless pathway. Shf is required for Hh stability and for lipid-modified Hh diffusion. PMID: 15691765
  4. Shifted is a secreted protein that acts over a long distance and is required for the normal accumulation of Hh protein and its movement in the wing. PMID: 15691766

Tissue Specificity
At the blastoderm stage, it is ubiquitously expressed. As embryogenesis continues, it is expressed in the epidermis and central nervous system, this expression being segmentally modulated. Also highly expressed at the foregut and hindgut throughout embryo
