Type VII collagen encoded by the COL7A1 gene is a crucial component for the epithelial-stromal attachment in skin, mucosa, and cornea. It consists of three identical alpha collagen chains, forming stable fibril structures that are key components of anchoring fibrils, ensuring the integrity and strength of tissues such as skin. Mutations in COL7A1 can lead to the absence or dysfunction of Type VII collagen, resulting in dystrophic epidermolysis bullosa (DEB). Although COL7A1 itself does not directly participate in signal transduction, as a part of the extracellular matrix, it may interact with cell surface receptors, indirectly affecting signaling pathways such as integrins and the transforming growth factor beta (TGF-β)/SMAD pathway, which play crucial roles in cellular biological processes.
