IDO1, indoleamine 2,3-dioxygenase 1, which is an enzyme encoded by the IDO1 gene, is mainly found in various tissues and cells outside the liver, such as the thymus, small intestine, epididymis, placenta, kidney, spleen, pancreas, and central nervous system, etc. The main mechanism of action of IDO1 is to catalyse the conversion of tryptophan to kynurenine, a process that is the rate-limiting step in tryptophan metabolism. In terms of signalling pathways, IDO1 activity is regulated by a variety of cytokines such as IFN-γ and TNF-α, which induce IDO1 expression by acting on the interferon-stimulated response element (GAS) and interferon-activated site (ISRE) of the IDO1 gene. Biologically, IDO1 plays an important role in the maintenance of immune homeostasis and regulation of immune response, especially in the prevention of foetal rejection during pregnancy, tumour immune escape, and in certain infectious diseases.Abnormal expression or activity of IDO1 is closely related to the development of various diseases, including tumours, autoimmune diseases and neurodegenerative diseases.

IHC image of CSB-RA215054A0HU diluted at 1:100 and staining in paraffin-embedded human endometrial cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
