KLK5 is a serine protease in the kininase family that plays multiple functions in the human body. It mainly participates in the physiological desquamation process of the skin and maintains the normal renewal of skin cells. However, the upregulation of KLK5 activity may trigger continuous skin desquamation, activate protease-activated receptor-2 PAR2, lead to the release of pro-inflammatory cytokines, and thus damage the epidermal barrier function, which is closely related to the development of inflammatory skin diseases. In addition, KLK5 may also participate in the immune regulation process of diseases such as rosacea by shearing Cathelicidin to produce the active form LL-37. Although its specific molecular structure and direct involvement in the signaling pathway need further study, the important role of KLK5 in skin health, cell proliferation, apoptosis, and immune regulation cannot be ignored.
