TNFSF15, also known as Vascular Endothelial Growth Inhibitor (VEGI) or TL1A, is a protein encoded by the TNFSF15 gene located on chromosome 9q32. This protein, composed of 251 amino acids, encompasses a cytoplasmic domain, a transmembrane domain, and an extracellular domain, and is capable of forming a stable homotrimeric structure. Primarily serving as a ligand for Death Receptor 3 (DR3), TNFSF15 participates in regulating the VEGF/VEGFR signaling pathway, affecting processes such as endothelial cell proliferation, apoptosis, and lymphangiogenesis. It plays a crucial role in angiogenesis, lymphangiogenesis, immune regulation, and other biological processes, and offers new therapeutic targets for pathological conditions involving increased vascular permeability, such as tumors and strokes.
